研究领域:
1. 血栓形成的氧化还原调控机制
(1)应用二硫键异构酶家族的基因敲除小鼠,确定对血小板和凝血系统发挥正性和负性作用的二硫键异构酶,解析该家族对动脉和静脉血栓形成的平衡调控及氧化还原信号网络(redox signaling network)。
(2)鉴定血小板和凝血系统的主要蛋白的关键性功能二硫键,应用巯基组学技术,研究静息与活化状态巯基-二硫键交换的动态过程及调控机制。
(3)研发靶向二硫键异构酶的抗血栓药物。
2.二硫键异构酶家族的生理作用
(1)应用二硫键异构酶家族的基因敲除小鼠,认识该家族在胚胎发育、骨髓造血、糖脂代谢、精子生成等基本生命过程中的关键作用。
(2)应用多种组学技术,研究二硫键异构酶家族在维持不同细胞内质网蛋白质稳态中的功能,鉴定其调控底物和作用的半胱氨酸位点。
3. 血浆激肽释放酶-激肽系统(内源性凝血系统)在固有免疫中的作用
研究该系统在固有免疫和脓毒症中的作用,建立新的治疗内毒素血症的靶点和干预途径。
